This was, as noted, a good video for beginners, and I appreciate that the artist clearly explained most of the decisions she made. However, a biodegradable glitter was mentioned in passing, with no further info as to when you'd use one over the other. Does that version still biodegrade despite being encased in resin?

Non-biodegradable glitters contain microplastics that pollute our environment but biodegradable glitter eliminates that problem by being made of cellulose. I think Twinkie uses biodegradable glitter to protect what DOESN'T make it into the project, not what does. But don't worry, your project is safe, it won't decompose in the resin.
